In Vitro Propagation of Ranunculus (Ranunculus asiaticus L.)





Abstract
Present experiments were carried out to examine the effects of explant sources, different basal media, various concentration of plant growth regulators on the direct somatic embryogenesis and histological characteristics of somatic embryo. Attempt was, also, made to examine the effect of various concentrations of plant growth regulators on the plantlet regeneration from somatic embryos.
The results obtained were summarized as follows:
Direct somatic embryogenesis was obtained from hypocotyl and cotyledon explants cultured on White¢¥s medium supplemented with 0.01§·/§¤ IAA and 3.0§·/§¤ BAP. The histological examination showed that initial cells had probably developed from sub-epidermal tissue of hypocotyl. Formation of secondary multiple embryos in the tissues of procmbryos were obsered on hypocotyl explant. Embryogenic cell divisions appeared to be initiated from epidermal tissue of cotyledon explant. In plantlet regeneration from somatic embryo, abundant roots were formed when IAA was used. MS medium supplemented with 1.0§·/§¤ GA©ý and 2.0§·/§¤ BAP were found most suitable for plantlet regeneration and bulblet formation.
